Proteins that work at the ends of DNA could provide cancer insight

ScienceDaily (Nov. 29, 2012) ? New insights into a protein complex that regulates the very tips of chromosomes could improve methods of screening anti-cancer drugs.

Led by bioengineering professor Sua Myong, the research group's findings are published in the journal Structure.

Myong's group focused on understanding the proteins that protect and regulate telomeres, segments of repeating DNA units that cap the ends of chromosomes. Telomeres protect the important gene-coding sections of DNA from loss or damage, the genetic equivalent of aglets -- the covering at the tips of shoelaces that keep the ends of the laces from unraveling or fraying.

Telomeres play an important role in cell aging and death, since each time a cell divides, a little bit is lost from the end of the telomere. Thus, cell biologists postulate that telomere length can determine the lifespan of a cell. Cancer cells, however, have a way to get around this limitation: An enzyme called telomerase that adds length to telomeres is highly active in cancer cells. This allows cancer cells to divide in perpetuity, running amok through tissues and systems.

"Cancer researchers want to get a hold of this problem, control this indefinite lengthening of the telomeres," said Myong, who also is affiliated with the Institute for Genomic Biology at the U. of I. "A lot of the anti-cancer drugs are targeted directly to these telomeres so that they can inhibit telomerase activity. The proteins we study regulate the activity of telomerase."

Using a technique developed at Illinois that allows researchers to watch single molecules interact in real time, Myong's group determined how two proteins called POT-1 and TTP-1 bind to the telomere. POT-1 protects the fragile telomere ends from being attacked by other regulatory proteins that might mistake the end for a broken or damaged area of DNA. When POT-1 and TTP-1 work together in a complex, they promote telomerase activity, an interesting target for cancer researchers.

The group found that on its own, POT-1 binds to the folded-up telomere in distinct steps at particular points in the telomere's DNA sequence, unfolding the telomere in a stepwise manner. However, the POT-1/TTP-1 complex surprised the researchers by binding, then freely sliding back and forth along the telomere end.

"Instead of stepwise binding, what we saw was a mobile protein complex, a dynamic sliding motion," Myong said. "Somehow it was as if the static binding activity of POT-1 is completely lost -- the protein complex just slid back and forth. We were able to reproduce the data and confirm it with many different tail lengths of the telomeric DNA and we know now that the contact between POT-1 and the telomere is somehow altered when the partner protein comes and binds."

Next, the researchers will add telomerase and see how the sliding activity of the POT-1/TTP-1 complex affects telomerase activity. Myong postulates that the sliding may promote telomerase activity -- and thus telomere lengthening -- by making the end of the telomere accessible for the telomerase enzyme to bind.

"We are excited about the possibility that this kind of mobility can increase the telomerase extension activity," Myong said. "It's somehow engaging the enzyme so that it can stay bound to the DNA longer. So it must involve a direct interaction."

Ultimately, understanding the POT-1/TTP-1 complex gives drug developers a new target for anti-cancer drugs, and the assay Myong's group used to monitor the complex could offer a venue for evaluating telomere-targeting drugs.

"We want to extend our a basic science knowledge in telomere biology into causes of cancer and we hope that our assay can be useful for telomere-targeted drug screening," Myong said.

Spending stumbles in Oct. as income growth stalls

U.S. consumer spending fell in October for the first time in five months as income growth stalled, suggesting slower economic growth in the fourth quarter.

The Commerce Department said on Friday consumer spending fell 0.2 percent after an unrevised 0.8 percent rise in September.

It said it could not quantify the impact of superstorm Sandy, but added it made adjustments where source data was not yet available or did not reflect the effects of the storm.

Though the storm, which slammed the East Coast in late October, put a brake on automobile sales, the drop in spending last month was in part a reflection of the weak economic fundamentals.

Economists polled by Reuters had expected consumer spending, which accounts for 70 percent of U.S. economic activity, would be flat last month.

When adjusted for inflation, consumer spending fell 0.3 percent, the first decline since June, after rising 0.4 percent the prior month.

It was also the largest decline since September 2009 and implied growth in consumer spending this quarter would struggle to exceed the third-quarter's 1.4 percent annual pace, which was the slowest in more than a year.

While the economy grew 2.7 percent in the third quarter after advancing 1.3 percent in the prior three months, much of the boost came from the restocking of goods and robust government spending. That is likely to be lost in the final three months of the year.

Growth could also be pressured by the lingering effects of the storm and automatic deep cuts to government spending and tax increases that could drain $600 billion early next year unless Congress and the Obama administration agree on a less-severe plan to cut budget deficits.

Income was unchanged in October for the first time since April and followed a 0.4 percent gain in September. The department said private wages and salaries fell, reflecting work interruptions caused by Sandy.

The amount of income at the disposal of households after inflation and taxes dipped 0.1 percent after being flat in September. Despite weak income growth, the saving rate rose to 3.4 percent from 3.3 percent the prior month.?

Scanadu unveils smartphone-enabled home diagnostics ...

scanadu SCOUTScanadu, the 2-year-old San Francisco startup aiming to develop a handheld medical scanner for home use, similar to the Star Trek tricorder, has unveiled its first three products, set for release by the end of 2013.

The Scanadu SCOUT is the central scanner component. The NASA Ames Research Center-based company says that customers simply hold the small, lightweight device to the patient?s temple and it will return five vital sign results with 99 percent accuracy in less than 10 seconds.

?Consumers? attention deficit is increasing,? CEO Walter de Brouwer told MobiHealthNews in an interview last month. ?And if they have a device that can do diagnostics, they are not willing to spend more than 10 seconds waiting. They?d like to have real time, they?ll settle for 10 seconds, but a minute would be too long.?

The data from the scan will be uploaded to a Scanadu app via Bluetooth and will track pulse transit time, heart rate, electrical heart activity, body temperature, heart rate variability, and blood oxygenation. The company plans to sell the device for less than $150, and the firmware will be remotely upgradable, allowing Scanadu to add new scanning metrics in the future.

?We found in user testing that the consumer doesn?t want to go through the ritual of the doctor checkup,? de Brouwer told MobiHealthNews. ?The consumer wants one position on the body. And he wants to do the diagnostics all in that one position. And the consumer is price-specific. For the consumer, the price sensitivity will be very high.?

In addition to the SCOUT, the company announced two ?low-cost and disposable early detection tools,? which will be integrated with the Scanadu app.

Project ScanaFlo will be used to conduct urinalysis. Customers can buy an over-the-counter disposable cartridge, and the software will test urine for preeclampsia, gestational diabetes, kidney failure and urinary tract infections. Pregnant women will also be able to scan for pregnancy complications or to monitor their health throughout the pregnancy.

Project ScanaFlu, meanwhile, will analyze saliva in cases of cold-like symptoms. It will test for Strep A, Influenza A and B, adenovirus, and RSV.

Scanadu plans to submit the products to the FDA for review as soon as next week. De Brouwer said that as a medical device with a consumer-focused use case, it?s not clear whether the FDA will consider the device unique. The SCOUT goes into manufacturing next month, freeing up the company?s team to work on the software side, particularly the user interface.

Scanadu also stressed in the announcement that Scanadu can be used by healthy customers, too, to monitor personal average vitals, so that when a person does get sick doctors have more information about what?s normal for that individual.

Dr. Alan Greene, Chief Medical Officer at Scanadu, said in a statement, ?Health decisions shouldn?t be based on averages, they should be based on a real, accurate and personalized healthfeed of data ? which we now have the power to give to the consumer in the palm of their hand.?

Scanadu is one of the more visible companies competing in the Qualcomm Tricorder X Prize, a competition to develop a device much like this one. The announced vitals the SCOUT can read line up fairly closely with the draft guidelines for the prize, with the exception of respiratory rate. De Brouwer said the device can read respiratory rate, but not within Scanadu?s self-imposed 10-second window. He said a marketable consumer product is a higher priority than the prize, which is still a year out from its first judging round.

De Brouwer believes Scanadu will be just one step in a personal health revolution.

?This is the beginning of a nascent industry of personal healthcare and consumer health. The first time in my life I?ve seen this excitement was with the personal computer, the second time was with the Internet and now it?s about health,? de Brouwer said.

He said he wants this product to change the relationship consumers of health care have with providers, making consumers the keepers of their own health information.

?Now we treat doctors as accountants who have to keep records for us,? he said. ?They were not trained to do this. What they were trained to be was an analyst of data, to give us their point of view, their therapy.?

WikiLeaks founder Julian Assange says all the necessary physical infrastructure for absolute totalitarianism through the internet is ready. He told RT that the question now is whether the turnkey process that already started will go all the way.

?RT: So you?ve written this book ?Cypherpunks. Freedom and the Future of the Internet? based on one of the programs that you?ve made for RT. In it, you say that the internet can enslave us. I don?t really get that, because the internet it?s a thing, it?s a soulless thing. Who are the actual enslavers behind it?

Julian Assange: The people who control the interception of the internet and, to some degree also, physically control the big data warehouses and the international fiber-optic lines. We all think of the internet as some kind of Platonic Realm where we can throw out ideas and communications and web pages and books and they exist somewhere out there. Actually, they exist on web servers in New York or Nairobi or Beijing, and information comes to us through satellite connections or through fiber-optic cables.

So whoever physically controls this controls the realm of our ideas and communications. And whoever is able to sit on those communications channels, can intercept entire nations, and that?s the new game in town, as far as state spying is concerned ? intercepting entire nations, not individuals.

RT: This sounds like a futuristic scenario, but you are saying that the future is already here.

JA: The US National Security Agency has been doing this for some 20-30 years. But it has now spread to mid-size nations, even Gaddafi?s Libya was employing the EAGLE system, which is produced by French company AMESYS, pushed there in 2009, advertised in its international documentation as a nationwide interception system.

So what?s happened over the last 10 years is the ever-decreasing cost of intercepting each individual now to the degree where it is cheaper to intercept every individual rather that it is to pick particular people to spy upon.

RT: And what?s the alternative, the sort of utopian alternative that you would put forward?

JA: The utopian alternative is to try and gain independence for the internet, for it to sort of declare independence versus the rest of the world. And that?s really quite important because if you think what is human civilization, what is it that makes it quintessentially human and civilized, it is our shared knowledge about how the world works, how we deal with each other, how we deal with the environment, which institutions are corrupt, which ones are good, what are the least dumb ways of doing things. And that intellectual knowledge is something that we are all putting on to the internet ? and so if we can try and decouple that from the brute nature of states and their cronies, then I think we really have hope for a global civilization.

If, on the other hand, the mere security guards, you know, the people who control the guns, are able to take control of our intellectual life, take control of all the ways in which we communicate to each other, then of course you can see how dreadful the outcome will be. Because it won?t happen to just one nation, it will happen to every nation at once. It is happening to every nation at once as far as spying is concerned, because now every nation is merging its society with internet infrastructure.

RT: And in what way are we, as sort of na?ve internet users, if you like (and I exclude you from that, obviously), kind of willingly collaborating with these collectors of personal data? You know, we all have a Facebook account, we all have telephones which can be tracked.

JA: Right. People think, well, yeah, I use Facebook, and maybe the FBI if they made a request, could come and get it, and everyone is much more aware of that because of Petraeus. But that?s not the problem. The problem is that all the time nearly everything people do on the internet is permanently recorded, every web search.

Do you know what you were thinking one year, two days, three months ago? No, you don?t know, but Google knows, it remembers.

So by just communicating to our friends, by emailing each other, by updating Facebook profiles, we are informing on our friends.

And friends don?t inform on friends. You know, the Stasi had a 10 per cent penetration of East German society, with up to 1 in 10 people being informants at some time in their life.

Now in countries that have the highest internet penetration, like Iceland, more than 80 per cent of people are on Facebook, informing about their friends. That information doesn?t [simply] go nowhere. It?s not kept in Iceland, it?s sent back into the US where it IS accessed by US intelligence and where it is given out to any friends or cronies of US intelligence ? hundreds of national security letters every day publicly declared and being issued by the US government.

RT: So do we risk kind of entering a scenario where there are almost two castes of people: a safe minority who are very savvy about the workings of the internet and the things that you described, and just people who go online for kicks?

JA: We have this position where as we know knowledge is power, and there?s a mass transfer as a result of literally billions of interceptions per day going from everyone, the average person, into the data vaults of state spying agencies for the big countries, and their cronies ? the corporations that help build them that infrastructure. Those groups are already powerful, that?s why they are able to build this infrastructure to intercept on everyone. So they are growing more powerful, concentrating the power in the hands of smaller and smaller groups of people at once, which isn?t necessarily bad, but it?s extremely dangerous once there is any sort of corruption occurring in the power. Because absolute power corrupts, and when it becomes corrupt, it can affect a lot of people very quickly.?

It?s just the matter of turning the key. And actually the key has already been turned a little bit, and it is now affecting people who are targeted for US drone strikes, organizations like WikiLeaks, national security reporters who are having their sources investigated. It is already partly turned, and the question is, will it go all the way?

RT: But has it been built really by corporations and kind of unwittingly subscribed to by people, in order to advertise products to make money, or has it been built deliberately by governments for the sole purpose of surveillance?

JA: It's both. I mean the surveillance infrastructure, the bulk surveillance infrastructure?? there are hundreds of companies involved in that business. They have secret international conferences, they have prospectuses that they give to intelligence agencies that we have obtained and published this year together with Privacy International and the Bureau of Investigative Journalism. Also, The Wall Street Journal has done some good work on this. They are building devices that they advertise to intercept entire nations, to install the data from those intercepts permanently ? strategic interception, because it's cheaper.

So it's a combined corporate/government amalgam. That's one of the problems, one of the reasons it's so unaccountable is that it crosses boundaries. Companies don't just sell to their home country, they sell to companies overseas. There are shareholdings held in BVI, and the company might be British-registered, like BIA, but actually a lot of research and development is done in Sweden, etc.

And then you also have Google and Facebook, who started up predominantly serving the public, but also have developed side projects to service the US intelligence complex. And individuals are constantly pushing their thoughts into Google as each thing that they want to research; it is pushed via emails, and on Facebook, through their social relationships. That's an undreamt of spy database.

Facebook is completely undreamt of even by the worst spying nation, given the richness and sophistication of relationships expressed.

RT: And willingly contributed to.

JA: Well, no. But not with informed consent. People don't actually know. When on Facebook it says "share this to your friends," that's what it says. It doesn't say "share this to state agencies," it doesn't say "share this to friends and cronies of state agencies."

RT: Who do you think has the organized power to stop these things that you are talking about?

JA: If there is political will, everything is possible. So if we get the political will, then of course those agencies can be dismantled. Very aggressive legislation, policing can be pushed upon them. In some regions of the world, such as Latin America, perhaps that's a possibility. There is a certain democratic tendency, which Ecuador is part of that might do that. But in general I think the prognosis is very grim. And we really are at this moment where it can go one way or the other way.

To a degree, perhaps the best we can be sure, if we work, of achieving is that some of us are protected. It may only be a high-tech elite, hopefully expanded a bit more ? people who can produce tools and information for others that they can use to protect themselves. It is not necessary that all of society is covered, all of society is protected. What's necessary is that the critical accountability components of society that stop it from going down the tubes entirely, that those people are protected. Those include corruption investigators, journalists, activists, and political parties. These have got to be protected. If they are not protected, then it's all lost.

RT: Is there a way that I can protect myself without knowing all about computers?

JA: Well, a little bit. But the first thing to be aware of is how much you are giving away. The first way to protect yourself is to go, "OK, I'll discuss that in person, and not over Facebook chat," or, "OK, I will discuss this using some forms of encrypted chat, like OTR, and not on a Facebook chat." You can go to torproject.org and download encrypted anonymizing software. It is slower than normal, but for things like internet chat it's fine, because you are not downloading very much at once. So there are ways of doing this.

What is really necessary, however, for those to be properly developed, there needs to be enough market demand. It's the same situation as soap and washing your hands. Once upon a time, before the bacterial theory of disease, before we understood that out there invisibly was all this bacteria that was trying to cause us harm?? just like mass state surveillance is out there invisible and trying to cause society a large harm.

?? no one bothered to wash their hands. First process was discovery; second process, education; third process, a market demand is created as a result of education, which means that experts can start to manufacture soap, and then people can buy and use it.

So this is where we are at now, which is we've got to create education amongst people, so there can be a market demand, so that others can be encouraged to produce easy-to-use cryptographic technology that is capable of protecting not everyone, but a significant number of people from mass state spying. And if we are not able to protect a significant number of people from mass state spying, then the basic democratic and civilian institutions that we are used to ? not in the West, I am no glorifier of the West, but in all societies ? are going to crumble away. They will crumble away, and they will do so all at once. And that's an extremely dangerous phenomenon.

It's not often where all the world goes down the tube all at once. Usually you have a few countries that are OK, and you can bootstrap civilization again from there.

RT: We just passed the second anniversary of Cablegate, and since then this war on whistleblowers and this state surveillance seems to have got worse. Do you think something as large as Cablegate could ever happen again and it would have a similar impact?

JA: Yes, yes. Hopefully next year.

RT: What sort of time next year?

JA: I won?t go into it, but hopefully earlier rather than later.

RT: Do you feel that when WikiLeaks is making these releases you?re having as large an impact as you?ve had before?

JA: Well, Cablegate was extraordinary. It was published over a period of 12 months. It?s the most significant leak. Our previous leak, on the Iraq war, was also 400,000 documents, showing precisely how over 100,000 people were killed. That was also very significant. But yes, no one has done anything as significant as that since, but yes, hopefully, that will continue.

The successes of WikiLeaks shouldn?t be viewed merely as a demonstration of our organization?s virility or the virility of the activist community on the internet. They are also a function of this hoarding of information by these national security [agencies]. The reason there was so much information to leak, the reason it could be leaked all at once is because they had hoarded so much. Why had they hoarded so much? Well, to gain extra power through knowledge. They wanted their own knowledge internally to be easily accessible to their people, to be searchable, so as much power could be extracted from it as possible. WikiLeaks attempts to redress the imbalance of power.

HTC confirms the Windows Phone 8S will not be coming to the US

The HTC Windows Phone 8S was announced as a lower-end alternative to its bigger brother, the flagship Windows Phone 8X, but the company has remained relatively quiet about it since. While we're still awaiting official details on pricing and availability across the globe, HTC has filled us in on one crucial nugget of information: it won't be coming to the United States. While we've been suspicious that this might be the case, we had been holding out hope that it might have at least some amount of reach in the US market. Essentially, HTC is focusing all of its stateside efforts on the 8X, its signature device. With that said, we'll still likely see the 8S show up as an unlocked and unsubsidized smartphone on various etailers, so Windows Phone enthusiasts with a penchant for bright colors will want to keep their eyes peeled. Now that we know more about the markets that won't be graced by the presence of the 8S, all we need now is a confirmation on when we can expect it elsewhere. Cancer drug shows promise in eradicating latent HIV infection

ScienceDaily (Nov. 29, 2012) ? Breakthrough drugs have made it possible for people to live with HIV longer than ever before, but more work must be done to actually cure the disease. One of the challenges researchers face involves fully eradicating the virus when it is latent in the body. A new report appearing in the December 2012 issue of the Journal of Leukocyte Biology suggests that a cancer drug, called JQ1, may be useful in purging latent HIV infection by activating the virus in the presence of potent therapy -- essentially a dead end for the virus.

"This drug may be useful as adjunctive therapy in efforts to purge latent HIV reservoirs to eradicate infection," said Monty A. Montano, Ph.D., principal investigator from the Section of Infectious Diseases at Boston University Medical Campus, in Boston, Mass. "This drug functions synergistically with other HIV purging agents."

To make this discovery, scientists used cell lines that contained latent HIV, as well as cells from patients who were on potent antiretroviral therapy with no detectable virus in their blood. Researchers then added JQ1 to latently infected cells, at physiologic concentrations, and observed potent reactivation of latent HIV. The researchers also observed potent suppression of inflammatory genes in genome-wide expression studies using the same cells. JQ1 reactivation of latent HIV may make it vulnerable to current therapies.

"One thing that's been made clear by the decades of HIV research is that there is no magic bullet for curing this disease," said John Wherry, Ph.D., Deputy Editor of the Journal of Leukocyte Biology. "Rather, the progress that's been made has been a series of incremental steps that often build on other previously developed therapies. The hope is that the ability of JQ1 to make latent HIV 'visible' to other HIV drugs described in this report will be another cog in the gearwheel of an HIV cure."
